Get 5 bottles of Bloodstained Tar Beast Blood from Bloodstained Tar Beasts at the Bloodstained Pond.
After you are done, you will find Ayesha right where you left her, by the Snoop Portal outside Boulderwind Village.
The map shows her standing in the middle of the Muddy Wheezers on the road between Mudpond Village and Boulderwind Village but this is false. She is not there.
